What is your leading hypothesis? That the etiology of schizophrenia is due to promoter hypermethylation and epigenetic dysregulation. The genes that are downregulated in key neurons of the brain are likely silenced because of epigentic factors related to promoter methylation. The observation that DNA methyltransferases are intimately associated with histone deacetylases provides a window into new protein targets that may prove to be amenable to pharmacological manipulation.
What piece of missing evidence would help prove it? Addional methylation studies in post-mortem brain samples, particularly from concordant and discordant twins.
